Is there any O/S named VRU?
Does it have anything to do with mainframes?

Many of the Voice Response Unit (VRU) systems interface with a mainframe, usually via a CICS transaction.

Nutcracker,

VRU = Voice Response Unit

Hardware or software, or both, that fields incoming calls by playing one or more prerecorded messages. The messages may require the caller to provide additional information by pressing buttons on a touch-tone telephone keypad. The sequence of messages played may be determined dynamically by this additional input.

I don't. The Telecomm world is a bit outside of my realm.

Logically, I would think that you would find information from the websites of the manufacturers of such equipment, who are probably the big players in the world of Telecomm (Periphonics, Lucent, MCI, Cicso, etc.). Maybe even IBM.
